The molecular and structural conditions underpinning presynaptic plasticity processes are less well understood than those involved in postsynaptic plasticity. We propose to address these questions by combining mouse genetics, organotypic hippocampal slice cultures, high-pressure freezing, freeze-substitution, and electron tomography. We will investigate and compare the 3D morphology of. Schaffer collateral and mossy fiber synapses, both at rest and during activity.
