Neural communication relies on neurotransmitter release and is therefore heavily influenced by the dynamics of synaptic vesicles. We generated a model of the presynaptic vesicle cycle, which combines experimental findings from several stages of the vesicle cycle and enables us to assess the dependence of vesicle release on synaptic input over multiple timescales, from milliseconds to hours. We now propose to build on this model and relate its parameters to the short-term dynamics of vesicle release, to understand how short-term plasticity arises, and how it relates to neuronal function.
