Cytoskeletal rearrangements and dynamics orchestrate several processes at the pre- and postsynaptic level. By benefitting from the recent developments of MINFLUX, a nanoscopy technique we invented, which is the first super-resolved optical method with true molecular-size resolution, we will elucidate the exact nanoscale organization of the synaptic actin cytoskeleton. We will shed light on the dynamics and molecular status of actin as a function of synaptic activity and strength by systematically analyzing the abundance and localization of proteins that regulate actin stability and dynamics.
